Most Alzheimer's disease (AD) research is directed towards studying �-amyloid plaques (A� plaques).  Recently, non-invasive positron-emission tomographic (PET) imaging has been used for detection of the formation of A� plaques using carbon-11 and fluorine-18 radiolabeled substrates. However the short half-lives of 11C (20 min) and 18F (110 min) may limit the widespread use of these substrates. Therefore, we have begun to evaluate novel AD substrates that can be radiolabeled with copper-64, which has a 12.7 h half-life and can be used for PET imaging. The current study involves a preliminary investigation of two novel bifunctional chelators (BFCs) that complex 64Cu and also bind to A� plaques. Two BFCs have been radiolabeled with 64Cu at good specific activities (258.0 and 305.6 Ci/mmol) and radiochemical purity above 95%. Results from serum stability studies and octanol/water partition coefficient [log P(oct)] values will be discussed for these compounds. In addition, initial biodistribution studies in normal mice will be discussed.
