With increasing development of bacteria with resistance to conventional antimicrobial agents, new silver based antimicrobials have been synthesized for direct nebulization into the lungs. Silver coupled to derivatives of caffeine-like xanthine compounds, silver N-heterocyclic carbene complexes (SCCs) or into shell crosslinked knedel-like nanoparticles (SCKs) is being investigated. The dose retention in the lungs and tissue distribution of silver loaded antimicrobial compounds in mice was studied using Ag-111 as a radiotracer. Ag-111 was produced from neutron irradiation of Pd targets and purified using ion-exchange chromatography. Pre-clinical studies using Ag-111 encapsulated in nanoparticles show increased nebulization efficacy with higher drug dosing and excellent lung retention (about 70 %ID/g) as compared with free 111Ag (I)[AgNO3 or Ag acetate]. The average %ID/g in the lungs was observed to be 50 times greater than most of the other organs studied�liver, spleen, kidneys and bone. Clearance of the compounds was mainly via fecal deposition.
