Norway spruce chips were subjected to SO2-ethanol-water (SEW) fractionation treatments to produce cellulosic pulps with controlled degree of polymerization and contents of lignin and hemicelluloses. The pulps were microfluidized and thin films of nanofibrillated lignocellulose (NFLC) were spin-coated on silica sensors to study the effects of lignin on enzyme activity using quartz crystal microbalance with dissipation monitoring (QCM-D). The QCM-D results show that enzyme adsorption and the hydrolysis rate decreased when the lignin content in the film increased. Interestingly, films with 1.4% lignin exhibited higher binding of cellulase mixture (celluclast) and cellulase monocomponent (CBHI) than films with 12% lignin which may be explained by the hydrophilic nature of SEW lignin. Complete hydrolysis of NFLC occurred within 20 minutes of enzyme incubation as revealed by atomic force microscopy.
