�-peptides and retro-inverse a-peptides represent two classes of peptide derivatives with great potential as foldamers for a wide range of biomedical applications. Here we present a new class of poly(retro-�-amide)s, termed �3r6 foldamers according to the nomenclature of �-peptide regioisomers (�3, �2, �2,3). Chiral building blocks were synthesized via both chiral pool approach and asymmetric synthesis, and efficient SPPS protocols were developed.The �3r6 backbone was designed to combine the topological distribution of inherent physicochemical properties of a- and �-peptides structures, and showed an interesting tendency to fold into �-sheet and �-hairpin whereas �-peptides tend to assume helical conformations.Besides �3r6 peptides with proteinaceous side chains, representative examples of �3r6 analogues of poly(amidoamine)s, polyhydroxy amphiphilic and zwitterionic polymers have been investigated. Due to the vast cross-disciplinary role covered by these classes of compounds and their range of physicochemical properties, �3r6 foldamers have high potential for chemical, biochemical and biomedical applications.
