Cotton fiber maturity is a major yield component and an important fiber quality trait that is directly linked to the quantity of cellulose deposited during the Secondary Cell Wall (SCW) biogenesis, and to the organization and orientation of crystalline microfibrils.  During the different phases of fiber development, important structural changes occur leading to cellulose macromolecules formation (b(1?4)D glucopyranose).  We have been using different techniques such as Fourier Transform Infrared Microspectroscopy and X-Rays diffraction to investigate the structure and organization of cellulose during different phases of fiber development from 10 until 56 dpa.  Two genotypes were selected for this study (TM-1 and TX55). FTIR results (both in reflection and transmission modes) revealed that this technique could be used as non-destructive analysis to document the changes occurring during fiber development. X-Rays diffraction analysis showed that the % crystallinity of cellulose increased from 20% at 17 dpa to 70% at 36 dpa. This drastic increase is followed by a drastic increase in cellulose content (increase from 20% at 17 dpa to 90% at 36 dpa).
