Beta-glucans are polysaccharides isolated from plant, fungal and bacterial sources and consist of diverse branching and linkage patterns of glucose building blocks that can act as triggers for innate immune responses. Unfortunately, the heterogeneity of naturally isolated samples makes the systematic investigation of structural variations on immune response arduous. Here we report the design of a library of various well-defined linear and branched glucan fragments to study the effect of chain length and branching pattern in innate immune responses. The synthesis of the library of di-, tri-, and tetrasaccharides using a unique solution-phase oligosaccharide synthesis automation platform and fluorous protecting groups will also be discussed.
