The study of the specific biological roles of human milk oligosaccharides (HMOs) has been limited by the availability of pure versions of the individual components. Herein, we report a strategy to chemically synthesize a library of complex HMOs using automated iterative solution-phase synthesis. The library was designed to make use of two versatile oxygen and sulfur-based fluorous protecting groups to simplify purification of intermediates.
