The chemical synthesis multivalent Pneumonia capsular polysaccharide conjugate vaccines are described. The vaccines consist of multivalent displays of the capsular polysaccharide minimum repeat structures from S. pneumonia serotypes 6B and 14 conjugated to a small molecule toll-like-receptor (TLR) agonist. Key steps of the synthesis include the use of our recently described Ph2SO/Tf2O/TBAI promoter system for iterative cis-1,2-a-glycoside synthesis to construct the serotype 6B minimum repeat unit, and the use of click chemistry to synthesize the multivalent constructs. The conjugates are currently undergoing biological evaluation for their ability to induce long-term immunity in animal models.
