RNA oligonucleotides containing a 2'-amino-5'-S-phosphorothiolate linkage (~2'-NH2-B1-ps-B2~) have potential application as substrates to investigate the mechanisms of ribozyme catalyzed reactions. Although the synthesis of dinucleotide (2'-NH2-U-ps-U) was reported previously, the synthesis of RNA oligonucleotide containing a 2'-NH2-B1-ps-B2 linkage remains challenging and has not been reported. By introducing the 2'-photocaged groups into the nucleoside phosphonate derivatives 1  and 2  , we have successfully developed a solid-phase synthesis protocol to prepare two RNAs containing a 2'-NH2-B1-ps-B2 linkage. The property of these modified RNAs and their application as a substrate in HDV ribozyme catalyzed reaction will be discussed.
