   Paracaseolide a, revealed as a valuable cell-cycle progression inhibitor with an IC50 value of 6.44 �M, was expeditiously synthesized in only eight steps from known compounds in 6.6% overall yield. The total synthesis features two sequential key transformations with a sulfenylation/oxidative elimination followed by a tandem 1,4-addition/aldol to stereoselectively functionalize the tetracyclic core. The versatile pathway tolerates multiple functional groups and provides facile access to libraries of diverse analogs.
