Abasic sites are the most prevalent lesion in genomic DNA.  We previously reported that the abasic site aldehyde can generate interstrand DNA-DNA cross-links via reaction with the exocyclic amino group of a guanine residue on the opposing strand in 5'-CAp sequences (J. Am. Chem. Soc. 2007, 129, 1852-1853). Here we will discuss the formation, stability, structure, and sequence requirements of Ap-derived cross-links in duplex DNA.
