The guadinomines are natural products isolated from Streptomyces sp. K01-0509. They are the most potent known inhibitors of the bacterial Type III Secretion System (TTSS). The TTSS is responsible for virulence of Gram-negative bacteria, including E. coli, Salmonella, Yersinia, and Chlamydia. The biosynthetic gene cluster of the guadinomines has been identified, and is a hybrid polyketide synthase/nonribosomal peptide synthetase. Insights into the function of the guadinomine synthetase will be described, as well as efforts to engineer new analogs of these promising anti-infective agents.
