Given the importance of small molecule therapeutics in the treatment of several disease classes, increased focus has been placed on the development of fast robust and flexible analytical tools for detection, isolation and characterization of new chemotypes. In this fast moving environment, the hyphenation of mass spectrometry to liquid separation techniques has become the most demanded analytical approach in the discovery of new therapeutic compounds. While straightforward in theory, for successful work within a production environment this approach requires the correct combination of orthogonal separation techniques to fully exploit the capabilities of mass spectrometry when dealing with complex matrixes. In this presentation, we will discuss the use of orthogonal separation and detection techniques to overcome the challenges associated with the identification and quantification of highly polar bioactive compounds (i.e. hydrophilic chemical degradation products and metabolites) from different sources. Details of new strategies for accurate compound detection by mass spectrometry will be presented, along with discussions of the strengths of powerful technologies like ultrahigh performance liquid chromatography (uHPLC) and supercritical fluid chromatography (SFC) in pharmaceutical drug discovery.
