Moniliformin is an important mycotoxin that has world-wide potential to contaminate cereal grains, including maize. Moniliformin contamination has been shown to cause death, reduced food intake, and weight loss in animals, which has led to the development of several traditional methods of detection for moniliformin. A new procedure for analysis of grains and feeds utilizing Ultra Performance Liquid Chromatography with Tandem Mass Spectrometry detection (UPLC-MS/MS) was developed and validated. Soybean seed (Glycine max) and maize grain (Zea mays) samples were extracted with aqueous acetonitrile. The UPLC column selected for the assay was a BEH Sheild RP18, 50 mm x 2.1 mm x 1.7 um. MS/MS detection provided excellent sensitivity and selectivity. The UPLC-MS/MS assay precision was 14.1% (relative standard deviation) and the overall mean spiking recovery was 88.6% across two sample matrices.
