Surfactants and detergents are important in today's protein biochemistry labs. They are required for solubility, stability, specificity and sensitivity in diagnostic assays. Since surfactants and detergents represent a diverse group of chemicals, assay techniques can be highly specific for any given surfactant. One of these detergents is sodium methyl oleoyl taurate, otherwise known as Hostapon� and other trade names. Sodium methyl oleoyl taurate is manufactured by a reaction of methyl tauride sodium salt with oleic fatty acid chloride. Commercial preparations of this material may contain low levels of unreacted materials that may result in precipitation of sample or high background issues when used in diagnostic assays. Two separate methods were developed for the detection and determination of these residual components in surfactant samples. Calibration curves were constructed using either purified oleic acid or N -methyltaurine (2-(methylamino)ethanesulfonic acid). Both methods include reversed-phase high performance liquid chromatography (RP-HPLC) and ultraviolet visible absorption spectrometry (UV-Vis). The oleic acid method utilized an YMC Pack ODS-AQ, 5 mm column. No preliminary derivatization step was needed for this method. HPLC separation was performed using an isocratic method with a mobile phase containing 95% acetonitrile, methanol, n-hexane and acetic acid (90/8/2/0.2 ratio) and 5% HPLC Water. The N -methyltaurine method utilizes a Waters AccQ-Tag, 4 mm column. Since N -methyltaurine has a low extinction coefficient, standards and samples underwent a pre-column derivatization reaction using 2,4-dinitrofluorobenzene (DNFB) to give a colorimetric derivative. HPLC separation was performed using an isocratic method with a 10 mM pH 6 Phosphate Buffer mobile phase.The proposed methods are accurate, reproducible and capable of detecting and quantifying the amount of residual oleic acid or N-methyltaurine in surfactant samples. These methods provide a simple screening approach for incoming lots of surfactants prior to their use in diagnostic assays or protocols.
