Cellular thiols play extraordinarily important roles in various aspects of cellular functions. Thiol redox state is considered to be the determining factor of the cellular redox state. Thiol density reflects the cellular oxidative state, especially thiol density on cell surface which reflects how cells respond to extracellular oxidative stress. Few methods are available to image thiol density on the cell surface in live cells through fluorescence microscopy. We would like to report a thiol specific fluorogenic agent that reacts specifically with various biological thiols to yield fluorescent thiol adducts with 370nm and 540nm as the excitation and emission wavelengths respectively. At the same time, the agent does not react with �NH2, -OH or COOH which are biologically relevant nucleophilic functional groups. Our data demonstrate that the agent effectively images cell surface thiols in live cells through fluorescence microscopy. Detailed experimental procedures and results will be presented.
