Investigations in our laboratories have focused on the synthesis and evaluation of various modafinil analogs as inhibitors of the dopamine transporter (DAT). Through previous studies, we have identified an effective inhibitor of dopamine transport that contains the diphenylmethyl aromatic subunit of modafinil tethered to an isopropylamide side-chain. In the current study, the isopropylamide side-chain of our known DAT inhibitor has been retained, while the aromatic portion of modafinil has been replaced with indole-based subunits. Synthetic and inhibition studies will be discussed.
