Variation in the structure of lignins originated from different lignocellulosic feedstocks is one of the major factors affecting the structure of final lignin products in various biorefining technologies. Therefore, understanding the structure of lignin in plant biomasses is crucial not only for fundamental aspects of lignin chemistry (such as lignin biosynthesis) but also for practical lignin applications.  Most of methods in lignin analysis require isolation of representative lignin preparations. This paper evaluates different analytical protocols for isolation of lignin and LCC preparations from various types of biomass (hardwood, softwood and non-wood materials). The study involves traditional lignin isolation procedures (MWL, CEL, acidolysis lignin), modifications of these methods as well as the use recently developed methods for biomass solubilization in lignin analysis. The advantages and limitations of specific preparations are discussed.
