Choline is a natural quaternary amine that has several important functions. It is a source of methyl groups needed to make the primary methyl donor, S-adenosylmethionine; a part of the neurotransmitter acetylcholine; and a component of the predominant phospholipids in membranes (phosphatidylcholine and sphingomyelin). The goal of this study was to determine the concentrations of choline-related metabolites in various tissues of rats using a stable isotope dilution-liquid chromatography-selective reaction monitoring-mass spectrometry (SID-LC/SRM/MS)-based assay that measures free choline, betaine, CDP-choline, phosphocholine, glycerophosphocholine,  phosphatidylcholine, sphingomyelin, trimethylamine N-oxide, methionine, S-adenosylmethionine and taurine. The assay was validated by assessing key parameters such as accuracy, reproducibility and repeatability, limit of detection (LOD) and limit of quantification (LOQ). Liver, kidney, brain, muscle, heart, and spleen were collected from three male Sprague-Dawley rats. The tissues were pulverized in liquid nitrogen prior to solvent extraction. Concentrations of the ten choline metabolites were compared among various tissue types and biological variations of the metabolites were also evaluated.
