The aim of this research is to develop a biosensor for the detection of clinical biomarkers. Towards this end, aptamers are used as molecular recognition scaffolds in conjunction with an enzyme (Glucose oxidase) co-factor as the signaling trigger. This particular study probes target recognition based signaling both via an indirect mechanism of enzyme (GOx) reactivation and a direct route via electrochemical detection of the co-factor. GOx catalyzes the oxidation of glucose into gluconic acid, however in the absence of its cofactor, flavin adenine dinucleotide (FAD) the enzyme can be rendered inactive. Ongoing studies include successful apo-enzyme preparation and reactivation via the co-factor. Additional steps towards electrochemical detection of co-factor have been taken to perform direct measurements of target binding. These studies will be used to design sensor strategies that utilize aptamer hairpin structures as target binding scaffolds that can signal the binding event via the co-factor.
