Reactions initiated and/or sustained by mechanical force (mechanochemical reactions) have evolved from a laboratory curiosity into a powerful alternative to conventional synthesis. [Fri�cic Chem. Soc. Rev. 2012 , 41,  3493.] The presentation will delineate the fundamental properties and generality of mechanochemical synthesis in a wide range of areas, including pharmaceutical materials, synthesis of existing and new potential medicinal targets, organometallic synthesis as well as porous metal-organic frameworks (MOFs). It will be highlighted how mechanochemical trasnformations provide a less solvent-intensive and more energy-efficient route to chemical synthesis from the simplest precursors, sometimes from raw materials that are not accessible to synthetic chemists due to their perceived inert nature and low solubility (e.g. mineral-like substances, such as metal oxides or sulfides). The results of the first methodology [Fri�cic et al. Nature Chem. 2013 , 5, 66.] for in situ X-ray diffraction monitoring of reactions during mechanical milling will be presented, demonstrating how real time mechanistic studies lead to a higher understanding of mechanochemistry and improve the ability to design mechanochemical processes involving inorganic, metal-organic (MOFs) or organic (pharmaceutical co-crystals) substances. Finally, a new, bioinspired technique of 'accelerated aging' will be presented, which mimics and accelerates the processes of biological mineralization (mineral neogenesis) to achieve a mild, solvent-free and low-energy route to conversion and separation of metal oxide or sulfide precursors.[Mottillo et al.
