A series of coumarin derived molecular probes show a ratiometric response to Co2+, Ni2+, Cu2+, and Zn2+ over other metal ions. The sensor showed a 65 nm hyposochromic shift in the UV-Vis spectrum (1:1 DMSO-TRIS, 10 mM pH 7.5), whereby the fluorescence spectrum showed a 20 nm bathochromic shift. The binding constants were calculated by the double reciprocal plot for each metal and found to be Log K11 = 4 and Log K12 = 10 : Co2+, Ni2+, Cu2+, and Zn2+, which are in agreement to the calculated values by HypSpec. Fluorescence microscopy experiments were carried out in ethanol solution to detect Cu2+ in biological media.
