A divalent Ruthenium poly-phenanthroline complex (Ru(NH2-phen)(Phen)2(PF6)) was linked to 5,10,15,20-Tetrakis-(4-carboxyphenyl)-21,23H-porphyrin (TCPP) by means of consensus sequences for protease detection. The resulting assemblies (after HPLC purification), as well as their components, were analysed by 1H and 13C-NMR, MS, UV-Vis absorption and fluorescence emission spectroscopy. Both, steady state and time-resolved fluorescence spectra of the assemblies were responsive to protease-induced cleavage of the consensus sequence between the ruthenium complex and TCPP. When detecting urokinase plasminogen activator (uPA), a protease activity down to 10-12 M could be detected.
