G-protein coupled receptors (GPCRs) are a family of cell membrane receptors essential for intercellular communication, demonstrated by their roles in smell, sight, hormonal regulation, blood clotting and neurotransmission. Of the ~900 human GPCRs cloned, 600 are termed �orphan� receptors since their extracellular agonists or ligands remain unidentified. GP133, an orphan GPCR, shows protein sequence similarity to four D. discoideum GPCRs that are activated by extracellular cAMP and regulate chemotaxis and cellular development, in part, through increasing intracellular cAMP. To determine whether GP133 plays a similar role in humans, the coding sequence of GP133 has been used to transfect COS-7 cells. Initial results suggest that treatment with extracellular cAMP results in a modest increase in intracellular cAMP similar to the D. discoideum receptors. These studies will be extended to show that the response is dependent on both extracellular cAMP concentration and the amount of DNA transfected.
