The focus of this project is to better understand the effects of metals and other oxidative conditions on DNA. This is accomplished by analyzing the amount of 8-oxoguanine (a compound known as an indicator of oxidative DNA damage) in damaged and undamaged cells. Analyzing the 8-oxoguanine concentrations gives a relative amount of oxidative DNA damage. The cells were grown in conditions that are known to cause oxidative DNA damage. Conditions include treatment with metal salts such as copper, iron, and vanadium. Peroxide and ultra-violet light are used to cause oxidation without the presence of metals. Cells were grown under the various conditions described above and the DNA extracted using a typical DNA extraction protocol. The DNA is then re-suspended in a buffer and analyzed using analytical techniques with the primary technique capillary electrophoresis. Ultimately this project determines if cell DNA damage can be quantitatively measured using novel instrumental techniques.
