Burkholderia cenocapacia, a bacterium that infects patients with cystic fibrosis, is highly resistant to many antibiotics. Its resistance to antibiotics is partly due to the presence of a lipopolysaccharide (LPS) layer located at the outer leaflet of the outer membrane (OM). The LPS layer decreases the permeability of the OM, preventing small molecules, including antibiotics, from entering the cell. In addition, the relative impermeability of the OM limits the ability to probe the components inside the LPS layer. Hence, we are using a transposon to generate mutations that cause increased permeability. The mutants with desired phenotypes are selected and the location of the transposon is identified.
