Oxidative stress is a cellular condition that has been implicated in various diseases such as cancer, Alzheimer's, Parkinson's, and Celiac disease. Cells under oxidative stress produce reactive oxygen species (ROS) that can covalently alter biomolecules. To aid in the analysis of oxidative stress, it would be useful to provoke cells to produce proteins on demand under oxidative conditions. However, the most common inducer of protein production, isopropyl �-D-1-thiogalactopyranoside (IPTG), has a sulfur atom that reacts under oxidative conditions. To remedy this complication, we turned to the C-glycoside mimic of IPTG, isobutyl-C-galactoside (IBCG). IBCG has been reported as a viable inducer, but its use under oxidative conditions has not been examined. In this study, we examined protein expression under oxidative conditions using a variety of inducers and compared their potency. In particular, we compared the induction efficiency of IBCG to that of IPTG and its products of oxidation. With an effective method to induce proteins under oxidative conditions, the study of oxidative stress and associated disease states will be facilitated.
