A series of fused heterocyclic (pyrimidine-2-thione, dihydropyran, pyrazoline, and valerolactam) derivatives of bis-benzylidenyl-4-phenylcyclohexanone and bis-benzylidenyl-N-methyl-4-piperidone were synthesized. The structures of the products were ascertained by spectroscopic studies and one was unequivocally ascertained by X-ray single crystallography. The target compounds showed modest cytotoxicity against murine L1210 lymphoma and B16 melanoma cells, with the most active derivative giving IC50 values of 7 and 15 �M for the two cell lines, respectively. The compounds did not cause microtubule depolymerization in cells, but several induced apoptosis in HeLa cells.
