Substituted chalcones have therapeutic potential, which includes anti-inflammatory, antibiotic, and anti-cancer properties. We have previously synthesized four chalcone isomers with nitro-substituents by novel methanesulfonic acid catalyzed Claisen-Schmidt condensation. The dinitrochalcone (DNC) products were characterized and evaluated for cytotoxicity in HEK-293T, MCF7, and MDA-MB-231 cell lines. The 4,4'- and 3,3'-dinitrochalcone exhibited low micromolar efficacies in inducing cellular toxicity in each of the cell lines tested. Interestingly, the 4, 3'- and 3, 4'- compounds show more varied results, with little or reduced biological activity in the tested assays. These results illustrate the bio-diversity of positioning of the substituted dinitrochalcones. We plan to test the DNCs to see if positioning of the nitro groups affects bioactivity in other assays as well.
