The ongoing analysis of the effects that electronegativity and steric hindrance have on the anticancer properties of non-natural isothiocyanates was continued with the synthesis of 12 halogenated benzyl isothiocyanates, a pair of benzyl ITC stereoisomers, and sulforaphane homologues.  Primary amines were converted into the respective ITCs, with appreciable yields, by the use of di(2-pyridyl)thionocarbonate.  Antiproliferation and chemoprevention assays against human MCF-7 breast cancer cells were conducted and provided insight toward the structural basis of isothiocyanates-dependent anticancer activity.
