Photodynamic therapy (PDT) has been used as a therapeutic technique to help localize and treat tumor target cells in tissues. The porphyrins interact with the target cells enabling them to be treated when exposed to the light. With this method in mind, Photodynamic antimicrobial chemotherapy (PACT) was devised to target bacterial cells instead of the tumor cells using the same treatment method as PDT. This study evaluates the mechanism of interaction between a bacterial target cell and the porphyrin sensitizer 5,10,15,20-tetrakis(methyl-4-pyridyl)-21H,23H-porphine-tetra-p-tosylate salt (TMPP) by examining the TMPP concentration before and after exposure. The study used both absorbance and emission spectroscopy in the quantification of the TMPP concentration on both Gram-positive and Gram-negative bacteria. Determining the interaction would support PACT and help improve its therapeutic methods of eliminating the target cells without harming the surroundings of the target cells.
