Peptoids are N-substituted glycine oligomers. They are attractive for study as peptidomimetics because they can form stable helical secondary structures and have enhanced biostability relative to peptides. In our laboratory, we are developing new techniques to study peptoid structure because high resolution techniques such as x-ray crystallography are difficult and expensive to use. In this project, we wish to understand tertiary structures of water-soluble peptoids. Limited information is available regarding peptoid tertiary-like structures. Here, we will use click chemistry to link two different peptoids, one of which we functionalize with an environmentally sensitive dye, 4DMN. Our putative tertiary structural model is a helix-turn-helix. Fluorescence and circular dichroism (CD) spectroscopic data are used to investigate intramolecular associations. Through these studies, we hope to clarify the sequence-structure relationships that are important for peptoid tertiary structure.
