Xanthene dyes like rhodamine B exist in equilibrium between an open, fluorescent state (1  ), and a closed, non-fluorescent state (2  ). It is common to evaluate new rhodamine derivatives based on different pH environments, but the polarity of the solvents used in an assay can greatly affect the open�closed equilibrium as well. We have used a polarity assay based on different dioxane/water solvent mixtures to compare the open�closed equilibrium of several rhodamine derivatives at constant pH and have found interesting trends based on different structural elements.
