Photoreceptor protein activation involves many decades of time, from photon absorption on the femtosecond timescale, the photochemical information storage events on the pico- to nanosecond timescale, to propagation of structural changes towards the molecular surface on micro- to millisecond timescales. Synchronized pairs of amplified Ti:sapphire lasers represent a powerful means to study these dynamic processes from the fs to ms timescales in great detail in a single experiment. Here, I will present time-resolved UV-vis, stimulated Raman and mid-IR spectroscopy results on a number of biological photoreceptors that are relevant in optogenetic and fluorescence imaging applications, such as phytochromes, LOV and BLUF domains and microbial rhodopsins.
