Molecular transport at the membrane of living biological cells has been examined in real-time observation conditions by a combination of Bright-Field Microscopy and the surface sensitive technique Second Harmonic Light Scattering. Studies of adsorption/transport of malachite-green (MG), crystal-violet (CV), and bromocresol purple (BCP), charged or neutral hydrophobic molecules, at Gram-negative and Gram-positive bacteria, and Murine erythroleukemia (MEL) cells have revealed the effects of a) membrane surface charge, lipid bilayer packing, and protein components; and b) the charge and polarity of the molecules; on the adsorption density and molecular transport rate. The studies have also revealed new understandings of the Gram-stan mechanisms.
