A native cyclofructan 6 (CF6) bonded silica column (Frulic N), a isopropyl carbamate-cyclofructan 6 column (Larhc P) and styrene divinyl benzene supported CF6 column (CF6-CMS) showed good separation capability in the hydrophilic interaction liquid chromatography (HILIC) mode. In this present work, the cytosine/uracil, benzyltrimethylammonium/cytosine, adenosine/adenine pairs were used to probe the hydrophilicity, cation exchange interactions, H-bonding interactions of the three CFs based stationary phases. Frulic N column exhibited the most hydrophilicity among the three columns and Larhc P column has less H-bonding interactions compared with Frulic N. The CF6-CMS was synthesized by clicking CF6 to chloromethyl modified styrene divinyl benzene resin so that the column displayed hydrophobic interaction due to the aromatic ring structure. In addition, the three CFs based stationary phases are found to be suitable for the separation of peptides, such as, cyclosporin A/cyclosporin C, oxytocin/octreotide/desmipressin, leuprorelin/gonadorelin/goserelin.
