We have recently rengineered the human Cellular Retinol Binding Protein II as a rhodopsin mimc to regulate the wavelength of bound retinylidene chromophore over 200 nm. A water network toward the end of the polyene plays a significant role in the wavelength regulation and pKa optimization of the rengineered protein by nearly 100 nm, and drops the pKa by four units. In this poster, spectroscopic and structurral data that allows us to investigate the mechanism of wavelength regulation will be presented.
