Based on the concept of biorefinery, triploid poplar(Populus tomentosa Carr.) was fractionated after the biopretreatment with the white rot fungi Trametes velutina D10149. The behaviors of hemicelluloses and lignin during the biorefinery process were elucidated in detail by both wet chemistry methods and spectroscopic analysis. The data indicated that Trametes velutina D10149 not specifically took the lignin components as the unique carbon source, simultaneously resulting in the biodegradation of carbohydrates. Differing from the gradual degradation of lignin bimolecular, sudden decrements of weight-average molecular weight (from 1.96�105 to 4.82�104 g/mol) and polydispersity (from 9.1 to 1.8) in the obtained hemicellulosic fraction after 4 weeks incubation were observed. The comparable analysis from both FT-IR and NMR spectra illustrated that the selective cleavages of certain linkages in hemicelluloses and lignin were not found by using Trametes velutina D10149. The basic and characteristic units were still detected in the hemicelluloses and lignin samples after 16 week incubation.
