Baker's yeast, Saccharomyces cerevisiae, is an established and commercially viable host for biopharmaceutical manufacturing. Yeast strains originally engineered by Novozymes Biopharma for the production of recombinant human albumin (Recombumin� Prime) have been used to express functional antibody fragments, such as monomeric scFvs, at multi-g/L yields in fed-batch fermenters. Genetic fusion of antibody fragments to human serum albumin can improve their pharmacokinetic properties, such as circulatory half-life, providing a reproducible and cost effective alternative to methodologies such as PEGylation. Protein engineering has improved the circulatory half-life of human albumin variants compared to wildtype albumin and sites have been introduced for the conjugation of bioactive compounds. A range of yeast strains and manufacturing process technologies have also been developed as a 'tool-box' for the rapid development of large-scale biopharmaceutical processes for the manufacture of antibody fragments and albumin fusion proteins.
