Peptides that fold and assemble into bundles of a-helices serve as models for natural protein domains and as building blocks for supramolecular materials. Conjugating synthetic polymers to these peptides is of interest to mitigate the physical properties of the peptides in applications ranging from drug delivery to light harvesting. Designing peptides that tolerate modification with synthetic prosthetic groups requires the delicate balance of interactions required for folding and assembly to be maintained. We are investigating the design and synthesis of peptides conjugated with monodisperse dendritic macromolecules to understand the interplay of how the structure of the dendron influences the folding and assembly of a bundle-forming peptide. This presentation will focus on challenges addressed during the synthesis of designed amino acid sequences in which several positions have been modified as sites for conjugation, and the successful conjugation of dendrons to these peptides.
