Oxygenation plays a critical role in health and medicine. Deficits or excesses in cells, tissues and organisms are associated with disease, damage, poor healing, and even death. To better understand normal physiology and disease states, it is advantageous to image oxygen in cells and tissues. Oxygen nanosensors provide increased combined spatial and temporal resolution over many traditional optical, electrode and other modalities. When fabricated as nanoparticles, iodide-substituted difluoroboron dibenzoylmethane-poly(lactic acid) (BF2dbm(I)PLA), with both fluorescence and heavy atom enhanced room-temperature phosphorescence (RTP), served as dynamic oxygen nanosensors for ratiometric imaging in tumor, brain and wound contexts. For increased tissue penetration of light and multiplexing applications, redshifted dyes with different emission wavelengths are desirable. Furthermore, ratiometric sensing benefits from distinct fluorescence and phosphorescence peaks with tunable and readily detectable intensities. To meet these goals, a series of heavy atom-substituted naphthyl boron dye-PLA conjugates were synthesized. Both bromide and iodide derivatives were explored. Optimal materials for ratiometric sensing based on Br-naphthyl-phenyl and iodo-dinaphthyl �-diketonate dyes were used to fabricate nanoparticles. The luminescence properties and oxygen sensitivity of the boron nanoparticles were investigated. Use of these materials in biological imaging studies will be discussed.
