To meet new demands such as higher titers and cleanability the evolution of Protein A media has been rapid the most recent years. The improvements have been both on the chromatography matrices and the Protein A affinity ligands. Today, binding capacity in the range of 60 to 70 g mAb per liter media is possible. This paper will discuss how the operating conditions can be optimized by a scientific approach using Design of Experiments, and how this will affect the capacity, yield and purity. In addition, productivity  calculations based on the different strategies  will be presented.
