Iron-sulfur clusters are found at the active sites of various metalloenzymes and are one of the main targets of nitric oxide (NO).  Upon NO interaction, dinitrosyl iron complexes (DNICs) can be generated. While the conversion of [Fe-S] clusters to DNICs by NO has gained attention for the past decade, the fate of the bridging sulfides during the conversion has been rarely discussed. We found the environment plays a significant role in the degradation of iron-sulfur clusters which was confirmed through our international collaboration supported by GREET.  The presence of acid or H� donors drastically alters the nature of the degradation products, which include the fate of the bridging sulfides.  The collaboration with Prof. Ivanovic-Bumazovic and Dr. Milos Filipovic at FAU in Germany allowed us to detect and confirm the generation of H2S from [Fe-S]/NO reaction using highly sensitive probes for H2S and NO.  Our results imply two gasotransmitters, NO and H2S, may be interconnected by [Fe-S] clusters. This talk will describe 1. the environmental factors that govern the reaction pathway, the reaction products' spectroscopic characterizations, and the plausible [Fe-S] degradation mechanism by NO and 2. the promotion of international collaboration at Brown University through the use of seminar talks and informal Q & A meetings.Reference1.
