In Protein A affinity chromatography, a wide range of elution buffer conditions are used in pilot and production scales.  Elution buffer choice can affect the characteristics of the elution pool and the subsequent downstream steps. This study explores the impact of a range of elution buffer conditions including molarity (0.01M-0.1M), pH (3.0-3.7), and buffer type (citric acid and acetic acid) on two different commercial Protein A resins (Eshmuno� A and Prosep� Ultra Plus). We monitor the antibody yield, elution pool profile, elution pool pH, and volume.  The effects of elution buffer concentration and pH on these parameters are established. Overall, citric acid seems to be superior to acetic acid as it generally provides more consistent elution pool characteristics. This could due to the fact that, compared with acetic acid, citric acid has higher buffering power and lower volatility. This work demonstrates the importance of the choice of Protein A elution buffer on the efficiency and process robustness of mAb purification.
