Past work in multimodal (MM) chromatography has focused on understanding the effects of various salts and fluid phase modifiers (FPMs) under low ionic strength conditions which are commonly employed in IEX chromatography.  In order to better probe the abilities of common FPMs to modulate hydrophobic interactions in MM systems, a reverse linear gradient elution screen was performed on a protein library and the results were compared to data from HIC systems. A variety of FPMs were used in the screen including arginine, urea and ethylene glycol. Analysis of the data showed that the addition of fluid phase modifiers in high ionic strength solutions produced qualitatively different results than when used in the IEX mode. This work sheds new light on how the strength of hydrophobic interactions can be controlled with FPMs in both HIC and MM systems.
