The solubility of sodium sulphate rises more than tenfold between 0� C and 32� C. Consequently its use in high concentrations in bioprocessing applications can be challenging. In large scale manufacturing facilities, buffers are subjected to variations in temperature and pressure that may adversely affect buffer stability. In an effort to mitigate this risk, a hydrophobic interaction chromatography process that originally required a high-concentration of sodium sulphate, was reevaluated with a lower sodium sulphate concentration in conjunction with a secondary electrolyte. In this case study, the HIC process for a recombinant protein was evaluated for optimum buffer components by testing two different concentrations of sodium sulphate in conjunction with six different concentrations of five other electrolytes. The entire study, including buffer preparation and sample purification, was automated on a Tecan Freedom Evo using GE PreDictor Plates, allowing all runs for the study to be successfully performed in duplicate in one business day.
