Real time, in-situ bioprocess analyte assays are of great interest in cell culture and microbial fermentation as means to improve process knowledge and understanding and to ultimately afford better feeding control.  Improved feeding control has been shown to be a primary, if not the primary, means to improved product quality and yield.  A new optical technology has been under development for in-situ, real time, automated closed loop feeding control.The new technology is optical but not spectroscopic and provides a global in-situ measurement of the metabolic state of the reactor and the kinetics associated with typical feeding protocols.  This system requires little a priori knowledge of the process, requires a simple single calibration process, and with its associated software suite provides powerful and effective closed loop control of feeding for cell culture and microbial processes.The hardware and software systems will be described along with theory and the operational protocol. Case studies will be presented and analyzed in cell culture and microbial applications.  The features and benefits of automated closed loop feeding control using this optical technology will be demonstrated and discussed.
