This study was undertaken to explore in vivo and in vitro anti-inflammatory potential of ssential oil obtained from leaves of Liriodendron tulipifera L (TLEO) and as well its mechanism of action in RAW 264.7 murine macrophages. TLEO was used to investigate lipopolysaccharide (LPS)-induced inflammatory responses in Raw 264.7 macrophages and inflammatory parameters such as cell viability, nitric oxide (NO) production, level of antioxidant enzymes (SOD, and GSH), TNF-a and reactive oxygen species (ROS), were evaluated. Topical application of TLEO significantly reduced ear inflammation in TPA-induced mice-edema model. Pre-incubation with TLEO significantly increased antioxidant enzymes with decreased cell toxicity in LPS challenged macrophages. On other hand, levels of pro-inflammatory mediators such as NO, TNF-a and ROS were reduced after treatment with TLEO as evaluated in LPS challenged macrophages. Evidences from these results, support that TLEO is a potent anti-inflammatory substance which could be used in therapeutic purposes.
