Malagashanine was isolated from the Madagascan shrub Strychnos Myrtoides, and its structure was assigned in 1994. It was found to exhibit chloroquine potentiating activity, and possess (along with several other strychnos alkaloids isolated form this species of shrub) previously unseen C3 relative stereochemistry. The development of methodology to address the new stereochemical pattern, and our subsequent approaches to malagashanine will be described. Development of the annulation methodology for application to structurally diverse classes of natural products will be discussed.
