Near-infrared (NIR) cellular imaging holds great potential in fluorescence-assisted studies of cancerous tissues. However, lack of stable NIR-fluorophores that can be activated by a user-designated reaction limits their use to provide real-time information on tumor cell microenvironment. Our turn-on-substrate-probe (TOSP) consists of a redox�active trigger group, a linker and an NIR fluorophore. Delineated here are the design and evaluation of this newly synthesized NIR-TOSP, whose fluorescent signature is revealed through probe activation by an enzymatic event occurring in tumor cells of multiple origins.
